The Spirulina takes one kind of the nutritious foods which widely uses by the people to know very well gradually.Therefore,to the research of Spirulina mainly concentrates on each active material now and includes in it,and in this experiment main research Spirulina polysaccharide this active material. It takes one pure natural material already to confirm that had the immunity adjustment, the anti-tumor, the anti-radiation, the anti-virus, the oxidation resistance, anti-senile and falls functions blood sugar and so on. But it is also unable to the Spirulina polysaccharide's extraction and the purification craft to achieve that the standardized operation to cause it to be unable by a more widespread useful. The present paper has carried on the optimization to the Spirulina polysaccharide's extraction's conventional hot water method, and has explored the Spirulina polysaccharide extraction one new method, and to improve the Spirulina polysaccharide the purification method to conduct the research. The result is as follows:The method extraction of Spirulina polysaccharide, this paper uses the conventional hot water extraction process to withdraw the spiral algae polysaccharidein the first time.Carried on the foliation about the extraction time, the liquid-solid has compared, the temperature and the extraction number of times carries on the single factor experiment to determine the technological conditions:extraction time 6h, the material fluid ratio 9:1, extraction temperature 80℃carries on two extractions for the constant temperature water bath. A new method to extract Spirulina polysaccharide has been developed which combines the ultrasonic extraction and cold lye extraction in this dissertation. Used the supersonic time, the extraction time, time the solution pH value and the extraction liquid-solid determines the good extraction condition compared to four factor three horizontal orthogonal experiments. The orthogonal experiment results show that the optimal extraction parameters with this method are:extraction room temperature, pH 9, the material fluid ratio 9:1, ultrasonic time 1 h, extraction time 2 h. The current method can improve the extraction yield of Spirulina polysaccharide with about 5 percent compared to the traditional one.In this paper it mainly carried on the extraction and the purification process the polysaccharideand the protein content comparison, and chose the method of sulfuric acid anthrone to carry on tests various stages total sugar content the determination, used the method of Folin-phenol to determine the protein content. Moreover, under optimum conditions of trypsin is carried out in four groups of pH 8 buffer solution, the determination of protease activity, determing in this experiment process the enzymolysis reaction time.In the Spirulina polysaccharide's purification stage, it has not used the gel chromatography and the highly effective liquid chromatography.But it has adopted method of simpler easy industrialization production. This paper divides into the main purification, the first auxiliary purification and second purifies three stages auxiliary, uses the trypsin enzymolysis, the kleit and the activated charcoal carry on the filtration as well as the dialysis process.The result proved that each purification stage has achieved the anticipated experimental result.After carrying on the determination, it is discovered that in the final product does not include the nucleic acid; The polysaccharide content above 98%, and the protein content is only about 1%.
