The leaves of Zanthoxylum bungeanum mainly located in the Province of Shaanxi, Gansu, Shandong , Sichuan. It tastes acrid and innocuous, and is edible. Our research is about the extraction,purification,antioxidant activity of the total flavonoids from leaves of Zanthoxylum bungeanum。The main research contents and results are as follows:1.Yield of total flavonoids is 3. 51% under the conditions Optimum conditions of ultrasonic-assisted extraction are: using the solvent of water, the temperature was 80℃, ratio of material to liquid 1:70, the ultrasonic power 360 W and the extraction time was 30min. The results show that optimum conditions of Ethanol solvent extraction are as follows: ethanol mass fraction 24%,optimal temperature for 70℃, ratio of material to liquid1:40, the extraction time was 25min, the ultrasonic power 360 W, yield of total flavonoids is 3.30% . Yield of total flavonoids is 3. 53% under Acetone: water (2:1) solvent.2.The optimum absorption conditions of macroporous resin D4020 are concentration of feed 2.19 mg/mL, pH value 5.And the the optimum desorption conditions are elute-ethanol concentration 70%, Twice the column volume of ethanol, adsorption and parsing options 2BV / h.After purification, the relative purity of flavonoids from 3.18% to 16.92%. While NaCl existed in the solution, equilibrium adsorption data fitted Freundlich iso-therms in the studied concentration range, as well as NaCl was inexistent. And the adsorption capacity in-creased when the content of NaCl rose.3. Extracting total flavonoids of water solvent scavenges DPPH ability for Ic50 24μg/mL, ethanol solvent for 17.5μg/mL, acetone solvent for 7.6μg/mL and Vc for 75μg/mL. The flavones had better reductive ability and scavenging DPPH ability that was better than Vc. The general trend : solvent of acetone extraction of total flavonoids > solvent of aqueous extraction of total flavonoids > solvent of ethanol extraction total flavonoids > Vc. The scavenging of the·OH radicals, overall trends is the ability to scavenging: Vc > solvent of acetone extraction of total flavonoids > solvent of aqueous extraction of total flavonoids > solvent of ethanol extraction flavonoids. 4. The flavones after purification scavenges DPPH? ability for 12.5μg/mL better then VC for 75μg/mL. The flavones had better reductive ability that was better than VC. The after purification total flavonoids of Zanthoxylum bungeanum leaves showed high anti-oxidative activity in vitro, which is a kind of valuable plant resources.The total flavonoids from leaves of Zanthoxylum bungeanum is a good natural antioxidants, which can be used as functional food, food antioxidants, healthy medicine.
