The technologies of extraction and purification of corn germ reduced glutathione were optimized which using corn germ as raw material, then studied the effect of antioxidation.the extraction reduced glutathione of Corn Germ as an index to determine the optimum extraction and broken method was ultrasonic.water formulation.Then, optimize the extraction condition of time, pH value and extraction temperature factors by using the Box-Behnken central composite design and response surface method (RSM) analysis of the establishment of the second regression model. Finally, we determine the optimum conditions of the water extraction of corn germ glutathione were as follows:extraction time was 19.6min, pH value was 3.7, extraction temperature was 91.7℃and the content of corn germ glutathione 0.145%. used column chromatography and solvent precipitation method to purificate reduced glutathione of corn germ.selected 732-type cation exchange resin column chromatography for purification by static adsorption and Analysis experiments. determined the best purification conditions of GSH by dynamic adsorption and analysis was as follows:The rate of adsorption amount of liquid and resin volume is 4; the adsorption time is 40min;the adsorption velocity is 1.0mL/min; the eluant was 0.2mol/L NaOH solution; the amount of eluant and resin volume ratio of 4:1. Selected ethanol as the best solvent of precipitation method, determined the optimum conditions of solvent precipitation method purificated GSH by single factors and orthogonal experiments were as follows:GSH extract and ethanol of 1:1volume ratio, the ethanol concentration is 100%, Stirring time is 15min.The content of reduced glutathione in Corn germ was purificated with the methods of column chromatography and solvent precipitation by dual-beam UV spectrophotometry, which respectively was 23.03% and 34.48%. This showed that the column chromatography purification as the best method. Improved the extract of the content of reduced glutathione significantly.Chemical methods was chosen to detect the extraction of GSH perared by resin initially.It was proved that the material of extraction contented GSH. Detected the substances of purification by Column chromatography through UV and HPLC, And the results was showed that the peak positions of purification was as similar as standard GSH. The stracture of purification was as similar as standard reduced glutathione through IR. Further evidenced that the material of extraction from the corn germ was reduced glutathione.Studied the antioxidant activity of reduced glutathione of corn germ and its antioxidant function in vegetables and fruits. The result indicated that the glutathione had the strong elimination ability to the free radical. Total antioxidant ability was 12.95U/mL, the maximum inhibition rate to hydroxy free radical was 80.19%,the maximum ability to Anti-superoxide anion was 135.2U/L,the maximum inhibition rate to DPPH was 79.37% and Anti-liposome was 49.62%. Reduced glutathione inhibited polyphenol-oxidase of vegetables and fruits, which could effectively delay the oxidation and brown of vegetable and fruit.
