Lily is widely physiologically active, containing various kinds of nutrient substances such as trace elements and vitamins, anti-cancer activity components like lily saponins and colchicine, and active ingredient with biological function like polysaccharides which can promote cellular immune function. The recent researches reveal that lily polysaccharides have the function of antitumor, hyperglycemic, oxidation resistance and anti-fatigue. In this paper, a general introduction of lily was given and recent researches on lily polysaccharides were summarized. Then, extraction of polysaccharides, purification and composition were studied, including the following sections.Firstly, polysaccharides were extracted by solvent extraction which was analyzed through the method of Response Surface. We had the best condition for extraction: temperature is 55℃; material-liquid ratio is 1:12; the time of extraction process is 3 hours; extracted for 2 times. The actual extraction rate of polysaccharide is 3.04% in this condition.Secondly, rough polysaccharides were isolated by DEAE 52-cellulose and three kinds of polysaccharides were gained:lily polysaccharides I was eluted by water, Lily polysaccharides II was eluted by 0.04mol/LNaCl; Lily polysaccharides III was eluted by 0.1mol/LNaOH. In order to get higher purified polysaccharides, Sephadex G-100 elution should be applied.Thirdly, the purified polysaccharide was identified roughly in the aspects of physical and chemical properties. Identification showed lily polysaccharide I was light yellow crystal, insoluble in cold water, ethanol, ether and other organic solvents, but slightly soluble in hot water. It contained uronic acid of 13.64% and didn't contain starch. Lily polysaccharide II was also light yellow crystal, insoluble in ethanol, ether and other organic solvents, slightly soluble in cool water, soluble in hot water. It contained uronic acid of 10.98% and didn't contain starch. lily polysaccharides III was light yellow powder, easily absorbing moisture, insoluble in ethanol, ether and other organic solvents, soluble in cold water and hot water. It contained uronic acid of 14.55% and starch.Fourthly, Polysaccharides' molecular weight was measured by HPLC. The results showed that the molecular weight of polysaccharides I was 97,000; the average molecular weight of polysaccharides II was range from 220000 to 465000; the average molecular weight of polysaccharides III was 94,000.Fifthly, the IR spectra applied to scan the lily polysaccharides showed that three kinds of polysaccharides had -OH, C=O, C-O-C and other polysaccharides absorption.The results of this study have important significance for compounded application of lily and improving lily's upgrade industries in Hunan.
