| Roses are considered to be very high medicinal value since ancient times. The modern medicine to be under the impression that roses have some efficacy such as strong lungs relief of boredom, stagnation and liver,, detoxification, to remove antibiotic-induced material injury endocrine balance, easing nerve fatigue, hangover etc. Polysaccharide is recognized that the physiological active substance with a specific health effect by. the food and medical profession.This paper mainly studies the aspects as follows:(1) To explore the crude rose polysaccharide bioactivity in Rose, we studied the best conditions and isolating techniques. Crude polysaccharide was extracted by hot water, then precipitation and decolorization by alcohol, and deproteinization by sevage method. The result shows that the optimal extraction condition is 90℃,4h and the ratio is 1:30 (w/v). The optimal extraction condition by microwave is 800W,4min and the ratio is 1:30 (w/v).(2) The crude polysaccharide was graded into two fractions:RP1 and RP2 by DEAE-52 chromatography, Sephacryl-400 and Sephacryl-200, which were shown to be homogeneous by HPLC, agarose gel electrophoresis and UV scanner.(3) Because RP2 occupies a large proportion in the two polysaccharides, therefore has the certain representativeness. Here we studied the structure of rose polysaccharides of RP2. The IR,1H NMR, 13C NMR and scanning electron microscopy analysis of RP2 showed that it is a lowly branched molecule. The results of our research suggested that RP2 was probablyβ-furan sugar.(4) The anti-oxidative function and anti-tumor activity of the polysaccharides were studied. The AP-TEMED reaction was used to produce and detect super-oxideanion (O2-·) and the H2O2/Fe reaction was used to produce and detect hydroxyl radicals(·OH).The results show that the scavenging ratio to·OH produced by H2O2/Fe system was 82.3%, to O2-·produced by AP-TEMED system was 87% under conditions of the experiment. The experimental result shows that rose polysaccharide has obvious antioxidant function to O2-·and OH·. The results show that rose polysaccharides have certain anti-tumor activity, the inhibition rate can reach the highest 61.27%. It can enhance immunity and antioxidant capacity so as to achieve the tumor Anti-bacterial. |
