In this study, the raw material was Osmanthus fragrans, we tried to use enzymatic hydrolysis technology to improve the extraction rate and quality of Osmanthus fragrans flavor, and used residue that after extracted Osmanthus fragrans flavor as raw material, prepared with flavonoids that had natural anti-oxidant activity, thereby have multitude of purposes, including full used of existing Osmanthus fragrans resources, processed enterprises to improve overall economic efficiency and reduced environmental pollution.By comparing three different enzymes on the yield of Osmanthus fragrans concrete, select theβ-glucosidase as the hydrolysis enzyme. Through single-factor experiment, the optimum conditions of Osmanthus fragrans concrete by insufflationβ-glucosidase hydrolysis technology were as follows: the dosage ofβ-glucosidase 0.5g/15g dry Osmanthus fragrans, hydrolysis time 2h, hydrolysis temperature 50℃, hydrolysis system pH5.0, the amount of aether 300mL, extraction temperature 40℃, under the optimum conditions, the yield of Osmanthus fragrans concrete byβ-glucosidase was up to 6.02%. Compared with conventional extraction method, sprayingβ-glucosidase hydrolysis method could hydrolysis fragrance precursor in Osmanthus fragrans, release flavor that the conventional extraction method is difficult to extract, not only the extraction rate of Osmanthus fragrans concrete was increased by 50% (the extraction rate of Osmanthus fragrans concrete by conventional extraction was 2.95%), the content of absolute of the Osmanthus fragrans concrete was increased, but also the content of effective volatile components was increased, thus achieving the desired intent of the experiment.Using the residue that after extracted the Osmanthus fragrans flavor as raw material, through the methods of conventional alcohol extraction and ultrasound-assisted extraction, extracted total flavonoids that have natural anti-oxidant activity in order to improve the efficiency of Osmanthus fragrans resource. By single factor experiments and orthogonal tests, determined the optimum extraction condition of conventional alcohol extraction method and ultrasound-assisted extraction method. They were as follows respectively: ethanol concentration 50%, extraction temperature 60℃, material/extraction solution ratio 1:80 and extraction time 2h; ultrasonic power 60W, ultrasonic extraction temperature 50℃, material/extraction solution ratio 1:80 and ultrasonic extraction time 30min. Compared with conventional ethanol extraction method, ultrasound-assisted extraction process was an ideal extraction method that had a higher rate of extraction, more process rapider, simpler, and had obvious advantages such as the production energy consumption. Based on the above experimental results of the reducing power, scavenging ability towards DPPH radical and lipid peroxidation inhibition assay can be drawn, the Osmanthus fragrans'total flavonoids extract had a better antioxidant activity in vitro, and its antioxidant activity in vitro was close to Vitamin C, in particular the ability of lipid peroxidation inhibition was far stronger than Vitamin C. So it had potential development value in the field of natural antioxidants after extracting Osmanthus fragran residue.Based on the above study, further isolate and purify the Osmanthus fragrans'total flavonoids extract, we firstly identificated preliminary structure of the Osmanthus fragrans'total flavonoids extract. By silica gel column chromatography separation to obtain three fractions, through discovery of its antioxidant properties were determined, fraction 1 and fraction 3 had btter antioxidant properties. LC-DAD-ESI/MS analysis showed that the fraction 1 was mainly ?avonol glycosides and the fraction 3 was mainly biflavone.
