Matrix Solid-Phase Dispersion Extraction (MSPD) has been widely applied to the pretreatment of solid. semi-solid and liquid samples, because the method is efficient, inexpensive and rapid, and the extraction, purification and separation can be performed in one step. The common dispersants in MSPD are diatomaceous earth, silica gel. alumina and C18. MSPD is similar to solid phase extraction (SPE). The MSPD has been widely applied to extracting drug residues in animal, vegetables, fruit and plants. To the best of our knowledge, there are few reports about the extraction of falvonoids by MSPD.Flavonoids are important chemical constituents in plants, most of which exist as glycoside or carbon sugar forms, and few of which exist as free forms. The falvonoids have prominent physiological activities such as anti-inflammation, liver-protection, anti-oxidation and anti-tumor. Therefore, preparation and determination of flavonoids was significant for the traditional Chinese medicine.In this thesis. the chemical structures. pharmacologic activities and extraction methods of flavonoids were introduced. Based on the previous researches, the MSPD was applied to the extraction of flavonoids from S. baicalensis Georgir and shaddock peel, respectively, the conditions of the MSPD were investigated and optimized.We applied the MSPD to extract flavonoids (baicalein. wogonin. baicalin) from S. baicalensis Georgir and the extract was analyzed by CZE. Firstly, the four parameters of MSPD. including dispersant. elution solvent. elution solvent volume and the ratio of sample to diatomaceous earth, were examined and optimized. Under the optimized conditions, contents of baicalein. wogonin and baicalin of S. baicalensis Georgir were 11.42-27.68mg/g.5.31-9.53mg/g and 62.02-101.32mg/g. respectively. The recovery ofr baicalein, wogonin and baicalin were 68.73%-84.86%,75.91%-91.3% and 80.76%-89.81%, respectively. In the research of naringin from shaddock peel, the content and recovery for the naringin were 23.0mg/g and 79.2%. The extraction yield was higher than that obtained by ultrasonic assistant extraction.The results indicate that the extraction yields were satisfactory for the extraction of flavonoids from the traditional Chinese medicine using the MSPD method. The proposed method was simple, efficient and rapid.
