The Emergency Early Warning System Research Against Chemical/Biological Attacks

The zebrafish is a kind of tropical freshwater fish, and it has been widely used in the studies of developmental biology and environmental toxicology. With the devel-oppment of genetic engineering techniques in recent years, the use of transgenic zeb-rafish as a monitoring system for environmental contaminants has become a research focus.The green-fluorescent protein(GFP) gene is a new reporter gene which can only be expressed in cells, and emit a green fluorescent without any substrates or cofactors. It has no speciality in species,tissues or location, and can be used in monitoring gene expression, signal transduction, co-transfection, transformation, protein trafficking and localization, cell separation and so on.In this paper, we successfully establish a poison detection system using trans-genic technology and green fluorescent protein(GFP). We selected the AHRDtk fragment and pFRMwg+plasmid vector to construct a new vector, which was trans-ferred into the Zebrafish via microinjection, after digestion, connection and PCR identification. Finally, we got the stable transgenic Zebrafish to detect poison through passage and screening continuously.
