The active ingredient in natural product is the important raw material for pharmaceuticals. Flavonoids could improve cardiovascular circulation, reduce myocardial oxygen consumption and prevent high blood pressure and so on. Its high-purity products are acclaimed in the international market. To meet the high-end market demand, and aim at the low productivity in the process of producing raw material medicine for pueraria lobata crude flavonoids, we studied the process of extraction and separation of total flavonoids from pueraria in this article.Both the methods of ethanol reflux and ultrasound-assisted are considered to extract total flavonoids from pueraria. A single factor and orthogonal experiments are considered to optimize the process conditions, and to compare the extraction rates of the two extraction methods. The results show that extraction rate of ethanol reflux is 81.50%, and extraction rate is 91.26% by ultrasound-assisted extraction, raising 11.98% over the former method.We also examined the purification effect of flavonoids by resin and extraction. With pueraria content and yield as the evaluating indicator, these two purification methods were compared. The optimum process of extraction method as following: an equal volume of n-butanol as extractant, pH value was adjusted to 2-3, get the total flavonoids content and yields of product were 71.49%, 90.52%. The optimization process of S-8 resin adsorption purification as follows: adsorbent dosage was ten times of flavonoids, 70% ethanol as eluant, flow rate was 1.5 BV / h, and the total flavonoid content and yields of the product were 82.45%, 93.02%. The results show that using resin adsorption purification of total flavonoids is the effective method, it is more suitable for industrial production.
