Pueraria lobata, i.e. Kudzu, is the perennial deciduous vine plant which belongs to pueraria category, leguminous family. Major components in Pueraria lobata are carbonhydrate, plant protein, amino acid, trace elements such as potassium, sodium, calcium, magnesium, zinc, and daidzin, flavonoids, isoflavones et al. Pueraria lobata have effective such as antipyretic, promoting the secretion of saliva or body fluid, promoting the positive aspect of body and the eruption, regulating body sugar level and protecting the vessels of heart and cerebral, its extractives are applied in food, health product and pharmaceuticals. There are abundant resources of Pueraria lobata in western Hunan region. In this paper, using wild Pueraria lobata from western Hunan region as the raw materials, with ultraphonic extraction, precipitation and adsorption separation, ultraviolet spectrum and atomic absorption analyze as separation ang analyze technology, separation technology of total flavonoids and total polyphenol in Pueraria lobata extractives were researched by calcium ion precipitation and resin adsorption, and its antioxygenation activity were investigated. The content of 5 trace elements were detected by flame atomic absorption spectrophotometry.With Pueraria lobata from Xiangxi as the research object and the extraction ratio of the total flavonoids as the response value, the influence of precipitation temperature,time and precipitator on the extraction ratio of the total flavonoids were researched with single-factor experiment, respectively. According to center united experiments design theory of the Box-benhnken, the separation technology of the total flavonoids in Pueraria lobata were optimized with response surface methodology(RSM) based on single-factor experiment. The conclusion showed that the extraction rate of the total flavonoids was 0.6589% in the condition of 50 m L extract, the precipitator of 70 m L saturation clarified limewater, the precipitation temperature were 60 ℃ and time was 45 minutes.It was well matched with the predicted value.With Pueraria lobata root as test raw material, adsorption separation technology of total polyphenol by macroporous resin was optimizated by response surface methodology on basis single-factor experiment. Influence of p H, adsorption temperature and time to extraction ratio of total polyphenols were investigated. Mathematic model wae established. Experimental results show that extraction ratio of total polyphenol is 0.2623% at 59℃ adsouption 180 min in p H 6.0. Predicted value of experimental model on extraction ratio was 0.2636%. Experimental results decreased 0.0013% in comparison with predicted content of model.The sample were digested with microwave for addition HNO3-HCl O4, and the contents of the Kalium, Sodium, Calcium, Magnesium and Zinc in root of Pueraria lobata were measured by flame atomic absorption spectrophotometry(FAAS). The results showed that contents of the Kalium, Sodium, Calcium, Magnesium and Zinc are 6.1638, 6.4451, 12.5357, 2.0142 and 0.0547(mg/g), repeatability, its recovery of adding standard sample are 98.89%, 97.30%, 101.01%, 96.81% and 96.12%, its relative standard deviation are 2.11%, 1.29%, 0.79%, 1.92% and 4.06%, repeatability. Relative standard deviation of precision and repeatability experiment are 0.65%, 0.29%, 1.01%, 0.57%, 1.01% and 0.26%, 0.61%, 0.43%, 0.22%, 1.35%, repeatability.The deoxidation ability and free radical scavenging ability of extractive from Pueraria lobata were investigated. The results showed that the peroxide value of the sesame oil which added Pueraria lobata extractive has decreased 7.84%, 21.42% and 15.56%, 59.30% in comparison with the added BHT and without added antioxidant under UV irradiation and heated at 60 ℃for 6 hours. Under these conditions, the deoxidation ability of Pueraria lobata extractive is 1.38 times that of BHT; The superoxide anion radical scavenging activities, H2O2 scavenging capacity and hydroxyl free radical scavenging ratio of the Pueraria lobata extractive at 50 ℃is 1.01 times, 1.02 times and 1.06 times of BHT, respectively.In this paper, the high-quality and rich kudzu resources were chosen as the research object, the determination of five metal elements beneficial to human body of content total polyphenol antioxidant activity were further studied based on the process of separation, purification and extraction. Theoretically support the development of the regional economy by rational develop and utilize the local kudzu resources.
