Docynia delavay is a specific wild fruit in Yunnan. It contains various nutrients and its healthy benefits have also been recognised. As we know that there is rare research on Docynia especially about bioactive components. Due to abundant nutriment, Docynia was considered potential hygienical food. There is no systemic report on the research of extraction and separation technique of total effective components of Docynia in the world. Our objection was comprehensive utilization prospect and exploitation of Docynia. With colorimetric method, quantitative detection of various effective components of Docynia delavayi, optimized the extractive paramenters of ultrasonic technique which was optimized by orthogonal design. The exchange behaviour of statie state and dynamic state of the D101 macroporous adsorption resin for the saponins in the Docynia delavayi were systematically explored. The effects of the elution requirement were also studied. As a exploitation, the mainly results are shown as follows:By preliminary analysis experiments, we proved that Docynia contain plenty of effective components, such as alkaloids, saponins, sugar, flavones, cardiac glycosides.Based on UV-VIS spectrophotometry, the content of total of saponins was quantified when it has been colored by vanillin acetum. The content of total of flavones was also determined by UV-VIS spectrophotometry.5% vanillin acetum and perchloric acid as color developer, pure oleanolic acid as reference, the content of oleanolic acid in Docynia delavayi were determined by spectrophotometry at 556 nm. The absorbency standard curve equation was A=67.73 C-0.002. Choosing 256 nm as identity wavelength of total flavones and using quercetin as a standard substance. The absorbency standard curve equation was A=89.46 C-0.007. Choosing phenol-sulfuric as color developer, glucose as reference, the content of total sugar in Docynia delavayi were determined by spectrophotometry at 448 nm. The absorbency standard curve equation was A=0.016 C+0.001. The contents of oleanolic acid, saponins, flavones, sugar were 0.169%,1.717%,1.478%,41% respectively. Ultrasonic extraction technology on total saponins and flavones from Docynia delavayi were performed. On the base of single factors experiments, the ultrasonic extraction techniques of Docynia delavayi saponins were optimized by orthogonal test. The statistical method disposed the results of test. The optimum extraction conditions were summed up as follows:the extracting time was 1 h, the solvent ethanol was 90%, the solid-liquid ration was 1:15 (g:mL). And the optimal parameters of extracting flavones as follows:the extracting time was 1 h, the solvent ethanol was 80%, the solid-liquid ration was 1:15 (g:mL).Via macroporous adsorption resin, the total of Docynia delavayi saponins were separated. D101 macroporous resin adsorption was a fit technique for purifying the saponins of Docynia delavayi. Seeing about the absorption and desorption capabilities between D101 macroporous adsorption resin and Docynia delavayi saponins, the optimul conditions as follows:pH of 3, the temperature was 25~35℃, the concentration of Docynia delavayi saponins was 6.45 mg/mL, the following rate of absorption and desorption were 0.53 mL/min and 1.06 mL/min respectively, the following solvent ethanol was 30%. The purification rate and recovery rate were 40.01%,54.9% respectively.The results suggested that the purification of total of Docynia delavayi saponins with D101 macroporous adsorption resin had been considered having antioxidant activity.The results of antioxidant activity showed that the content of saponins of scavenging 50% hydroxyl radical (CC50) was 0.1569 mg/ml, the content of saponins of scavenging 50 % superoxide free radical (CC50) was 0.8685 mg/ml.In a word, to synthesis the result of the whole research, we found these techniques such as ultrasonic extraction and D101 macroporous adsorption resin.
