Wheat germ is the nutrition essence of the whole wheat. In this project, Wheat germ was chosen as the raw material, from which wheat germ oil with nutrition and health-keeping value and natural vitamin E were extracted by solvent extraction method. The topic mainly studied the optimal extraction condition of wheat germ oil and natural vitamin E. Various experimental factors impacting the effects were discussed and the main ones were chosen to Parameter analysis and optimization. In the end, the optimal extraction conditions of wheat germ oil and natural vitamin E were determined. Then wheat germ oil was refined after a series of operations and fully met the national index of edible oils standards.The main research contents and results summarized as following:(1) The topic mainly studied the optimal extraction condition of wheat germ oil. Several experimental factors impacting the results were discussed, including extractive solvent, proportion, Particle degree, water content, time, temperature, proportion between material and solvent and so on. Experimental results showed that the optimum conditions were the proportion between hexane and acetone 1/1, Particle degree more than 20 mesh, water content 2%, time 4h, temperature 50℃, proportion between material and solvent 1/5.(2) The topic mainly studied the optimal extraction condition of natural vitamin E. Several experimental factors impacting the results were discussed, which were extractive solvent, water content, temperature, time, ethanol concentration, proportion between material and solvent and so on. Experimental results showed that the optimum conditions were ethanol concentration 95%, water content 4%, temperature 70℃, time 120min, proportion between material and solvent 1/4.(3) Wheat germ oil was refined in the methods of split step alkali. The topic mainly studied the conditions of degumming, Alkali refining, washing, decoloring and deodorization, then finding the optimal refining condition. Experimental results showed that using phosphoric acid enzymatic degumming method could effectively remove the glue component of wheat germ oil. The first adding alkali amount was 55 percent to 70 percent; the optimal temperature of decoloring and deodorization were 90℃and 180℃~200℃.(4) The residual solvents of wheat germ oil were tested using gas chromatography. Experimental results showed the residues of acetone and hexane in Original wheat germ oil were 22.7mg/kg and 41.2mg/kg, which didn't conform to National level of cooking oil standards. So it is necessary to remove residual solvents. Experimental results showed acetone was not detected and the residues of hexane in refined wheat germ oil was 7.5mg/kg, which conformed to National level of cooking oil standards.
