Macroporous adsorption resin method was firstly applied in decoloring of industry and separation of antibiotics, its application in separation and purification of active ingredient in natural product had started from the 1970s. Through researches of many years, at present macroporous adsorption resin method has been widely applied in the food industry and separating active principles of traditional Chinese medicine, etc. Nevertheless, we need to study how to realize the high-efficiency separation of active principles in natural product by macroporous resin.The study aimed at completing the high-efficiency separation of active principles in natural product by macroporous resin, macroporous adsorption resin mixed bed was firstly adopted to investigate the separation and purification of substrate, in which process, stevia crude extract, market steviosides and Lamiophlomis rotata crude extract were used as adsorbates, respectively. High performance liquid chromatograph (HPLC) and UV-VIS spectrophotometer were employed as examine devices. After studying the adsorption and desorption of dozens of macroporous adsorption resin, the separation and purification effect of macroporous adsorption resin mixed bed to different adsorbate was investigated, and obtained the following results:1. The ultrasonic method was applied to extract the total flavonoids from Lamiophlomis rotate, the related extraction conditions was optimized. Based on this, the separation and purification of total flavonoids form the extracts by macroporous adsorption resin mixed bed was probed, and obtained mixed bed of LZ-9 and LZ-48 with the best purification effect, the optimal extraction conditions of adsorption and desorption were: extract time tt = 180 min, extract temperature Tt = 45℃, ultrasonic intensity W = 60 %, extractant V_乙醇:V_水= 25 %, extraction times n = 4.2. The market steviosides was used as adsorbate, after the adsorption and desorption experiment of which on more than thirty macroporous resin, the macroporous resin mixed bed of LZ-1, LZ-20, LZ-30, LZ-37 and LZ-36 displayed the best separation effect for RA, the adsorption process also accorded with pseudo-second order reaction kinetics and Brunauer IV equilibrium adsorption isothem, and the rate-limiting step was intra-particle diffusion.3. Stevia crude extract was used as adsorbate, after the adsorption and desorption experiment of which on nineteen macroporous adsorption resins, the separation effects of Rebaudioside A (RA) from steviosides by macroporous adsorption resin mixed bed were investigated, and the related adsorption curves was obtained, furthermore, the conditions of separation RA rom steviosides by macroporous adsorption resin mixed bed were optimized. The results of the study indicated that the mixed bed consisted of LZ-31, LZ-21, LZ-20 and LZ-42 showed the best effect in separating RA from steviosides, the adsorption process accorded with pseudo-second order reaction kinetics, the isothem corresponded to the Brunauer IV equilibrium adsorption isothem.
