Mimosa contains flavonoids, phenols, biological polysaccharide, amino acid, organic acids, etc. These active substances beneficial to human body. The sensitive plant flavonoids has significant physiological activity, therefore it is meaningful to separation of the sensitive plant flavonoids. Macroporous adsorptio n resin is a new separation and purification technology, has several advantagess :remove impurities in the sample, purification target composition, has a wide application range, narrow liquid dosage, decrease product absorption of moisture, etc, in recent years has gradually been applied in the separation and enrichment of traditional C hinese medicine composition. The purpose of this study is that the macroporous adsorption resin separation technology is applied to the separation and purification of flavonoids in the sensitive plant.Chapter 1: the flavonoids and its measuring method are introduced, and the macroporous adsorption resin and macroporous adsorption resin purification technology are introduced and macroporous adsorption resin in separation and purification, the application of flavonoids.Chapter 2: soxhlet extraction method was optimized extraction process conditions of total flavonoids in the sensitive plant. To obtain rate and total flavonoids of flavonoid yield two indicators to measure and obtain sensitive plant the best extraction conditions: the solvent is 50% ethanol solution, extraction time was 2.5 h, best extraction solvent on the relative to the sample quantity is enough, the higher extraction efficiency.Chapter 3: A method for separation and purification of flavonoids from mimosa pudica by using Diaion HP–20 macroporous adsorption resin was studied. By using mimosa pudica product from Hainan and with the absorb amount and recovery rate of flavonoids as the evaluation index,the sample flow rate,the kind and amount of desorption solvent,diameter and heigh ration,elution velocity and desorption ration were evaluated. The optimum process conditions: the sample rate was 3 m L/min,eluent was 40% ethanol solution,column diameter ratio was 1∶10,and the elution velocity was 4 m L/min. The method is simple,feasible,and it can be used for effective separation and purification of flavonoids in mimosa pudica where the macroporous adsorption resin can be regenerated with high utilization.
