Titania, because of its chemical stability and its specific adsorption of phosphoprotein and phosphopeptide, was deemed to be a kind of emerging and good High Performance Liquid Chromatography packing material. Therefor, combined with titania's good prospects and the advantages of monoliths, developing titania monoliths column, and will be able to quickly and large quantities analyze, especially in proteins, peptides field. In this paper, synthesis of titania monoliths and the application of titania monoliths column in separation of phenol, commercial tiania column in separation of whey protein and its hydrolysate were studied.The researches in this thesis included three aspects:(1) Through an one-step synthesis of macro-mesoporosity bimodal titania monoliths via a template free sol-gel process, using titanium isopropoxide as titania source, economical and convenient acetate and hydrochloric acid as additives. The morphology of the synthesized titania monoliths can be controlled by adjusting the content of acetate and water in the initial reaction solvent. Titania monoliths with more than 15 cm in length, macropore of 1μm, mesopore of 5 nm, BET surface area of 63.4 m2/g, and Langmuir surface area of 118 m2/g was obtained after 350℃calcination, and used as high performance liquid chromatography column.(2)Using synthsised titania monoliths column analysis phenolic standards, and evaluate the performance of titania monoliths column by retention time, thermodynamic parameters.(3) Using commercial titania column separate whey protein hydrolysate. Phosphate buffer by different concentrations and different concentrations of acetonitrile, the best HPLC conditions: mobile phase 85% ACN, salt, sodium phosphate buffer pH5.0, salt concentration of 85% ACN conditions pH5.0, the maximum solubility of sodium phosphate 1.0mM.
