Polyrotaxanes (PR) as an important member of supramolecular chemistry, has special movable "collar" structure.As the "rotor" of the cyclodextrin (CD) ring can slip and rotate alonging the PR "axis"Its unique supramolecular structure for the separation of science provides a new way of thinking. But so far, the study on the polycyclodextrin based rotaxanes as the spacer of affinity adsorption material is little. In this paper, By click chemistry considered polycyclodextrins and rotaxanes as spacer of affinity adsorption materials and setted up a connection bridge between the ligands and the basement membrane.Futher,builded the new affinity membrane. The relationship between structure and performance were exploration. In biological macromolecular separation and purification field provides a potential method.This paper proposes to triazine dyes as biological affinity ligands.Cyclodextrins and rotaxanes (PR) as the spacer.Ethylene vinyl alcohol copolymer (EVAL) as a kind of affinity matrix material.A variety of methods, such as filling, chemical, and so on, are used to design and prepare a plate and hollow fiber affinity membrane,The static and dynamic adsorption of biomolecules on the membrane were studied, and the separation and purification of biological system was carried out.The new type of affinity adsorption material achieved good effect. Specific research contents and results are as follows:(1) A novel affinity membrane was prepared by blending, and the structure and adsorption behavior of the membrane were investigated. The results showed that the affinity membrane is asymmetric finger pore structure and the function body within the membrane distribution is relatively uniform. Affinity membrane has good adsorption capacity for BSA and other proteins. The affinity membrane for the three adsorption and desorption process, still has a high adsorption effect, and the affinity of the membrane is high repetition rate.(2) Click chemistry for sealing side reaction, β-cyclodextrin as the end capping agent, with one step by using water as a solvent, synthesis of the CD as the main body, PEG as the object of polyrotaxanes.The reaction conditions are mild and the operation is simple. The successful synthesis of the products was verified by NMR, IR and TEM.The results show that the PR of each PEG main chain contained 26 cyclodextrin molecules.(3) The preparation of the "can be clicked" interval arm CD and PR, And the CD and PR coupling to active carrier EVAL-alk with terminal alkyne. EVAL-PR and EVAL-CD affinity adsorption materials were prepared with different supramolecular spacer.The successful preparation of affinity membrane was verified by IR, XPS and ’H NMR spectra;The microstructure was characterized by SEM, which showed that the affinity membrane had a well-developed pore structure,and could provide a channel for the material liquid. At the same time, the static adsorption property of EVAL-PR was studied by fluorescence confocal microscopy and electrophoresis.The affinity membrane has good adsorption effect on lysozyme.(4) Preliminary hollow fiber affinity membranes were investigated.Studied the function body CD and EVAL blend hollow fiber affinity membrane.Meanwhile, the adsorption and desorption performance has been systematically investigated(i.e., initial concentration of lysozyme, medium pH, ionic strength), and the results showed that the affinity membranes have excellent adsorption performance. The phosphate buffer solution with pH=8.0 1mol/LKSCN was effective, and the desorption rate reached 90%.
