Wool keratin is an environmentally material that has good biological compatibility and good biodegradable. In this paper, the reduction system composed of multifunctional solvents and the single solvent TCEP were applied to dissolve wool keratin fiber to prepare keratin respectively. Then reduction keratin was mixed with sodium alginate and polyacrylonitrile, drug loaded wool keratin/sodium alginate film and wool keratin/polyacrylonitrile electrospinning film were prepared,respectively.The structure and properties of the restore C keratin and TCEP keratin were studied. Scanning electron microscopy (SEM) test showed that the scales of the wool was to be destroyed at first, then the solvent begin to dissolve the internal structure and chemical bonds of the wool. FTIR test showed that the wool and two kinds of protein powder containing â…  amide, â…¡ amide and III amide. DSC showed that compared with the wool, the bound water makes the endothermic peak of keratin to high temperature, keratin alpha crystallization melting peaks move to low temperature. Electrophoresis spectra test showed that the reduction keratin molecular weight is contained the higher molecules, but broad molecular weight distribution, the TCEP keratin molecular weight is more concentrated, but the molecular weight is lower.Wool keratin/sodium alginate flat membrane was prepared, and the effect of sodium alginate was studied by FTIR, drug-polymer interactions, the encapsulation rate. FTIR test showed that sodium alginate and keratin produced hydrogen bonding keyed and electrostatic forces, different concentration of sodium chloride and urea on the viscosity of blend solution showed that the electrostatic forces is more important. Swelling rate determination showed that with the sodium alginate increased, the encapsulation rate increases gradually, the drug-polymer interactions showed a trend of increase after decreases first, crosslinking agent calcium chloride select 8wt% in mass fraction. Mechanics performance test showed that the mechanical properties of the composite membrane were improved with the increase of sodium alginate content. Drug release performance test showed that with the increase of sodium alginate content, the control drug release ability was improved.The drug-loading of wool keratin/polyacrylonitrile electrospinning membrane was prepared and its properties were studied. Results showed that with the increase of keratin content, the blending of electrospinning fluid viscosity decreased. SEM test showed that with the increase of keratin in the blank wool keratin/polyacrylonitrile in electrospinning film, the fiber diameter decreased, the blank was smooth. Compared with the blank film, drug-loading wool keratin/polyacrylonitrile electrospinning film morphology compared the blank, fiber diameter is higher, fiber diameter distribution is wider. Contact Angle and slow-releasing experiment showed that the introduction of keratin has a certain influence on drug release.
