As a most widely used separation and detection method, HPLC has the advantage of rapid and efficient for the complex real samples with higher sensitivity and lower LOD. And the analytical method has the expansive function in the fields of food and drug analysis. Du to not all analytes possess fluorescence, the direct fluorescence detection is not suitable. Hence, the application of HPLC is vastly limited. Fluorescent derivative method as one of the most common supplementary means could perfect the application of HPLC. DLLME as an emerging technique has the advantages of simple and rapid operations, significant timesaving, low consumption of chemical reagents, good recovery and high enrichment factor. In this paper, investigations of food safty and medicine bioavailability were explored by using fluorescence labeling reagents which synthesized in our own laboratory combined HPLC and DLLME. The specific content as follows:Chapter one: HPLC, chemical derivatization, DLLME are introduced, and food safe and medicine bioavailability are summarized.Chapter two: a new method was developed for the determination of four nitrosamines(NAs) in foodstuffs based on fluorescence labeling followed by high-performance liquid chromatography with fluorescence detection(HPLC-FLD), which did not require the complex pretreatments and expensive equipment. NAs were firstly denitrosated to obtain the secondary amines. Subsequently, fluorescence labeling procedure was immediately carried out to label the secondary amines using 1,2-benzo-3,4-dihydrocarbazole-9-ethyl chloroformate. The proposed method was successfully applied for the determination of trace NAs in several foodstuffs, and provides a potential choice for the quality control of foodstuffs.Chapter three: a analytical method for ractopamine(RAC) screening in various kinds of foodstuffs based on fluorescent labeling coupled with DLLME was established by HPLC-FLD. And 2-(11H-benzo[a]carbazol-11-yl) ethyl chloroformate as an excellent fluorescent reagent was employed to label RAC, ensuring highly sensitive detection.Chapter four: 2-(12-benzo[b]acridin-5-(12H)-yl)-acetohydrazide as the fluorescence labeling reagent coupled with HPLC-FLD was developed for simultaneously detect multiple triterpenic acids in the presence of water. The proposed method was successfully applied to the study of triterpenic acid bioavailability with excellent applicability and good reproducibility.Chapter five: a novel HPLC-FLD method coupled with pre-column labeling using 2-(5-Benzoacridine) ethyl-p-toluenesulfonate as a fluorescent reagent for the determination of L-theanine in mice plasma was established and validated,and the bioavailability of L-theanine was investigated. The method was proved simple and timesaving.
