Acrylamide is a small molecule which has neurological toxicity and reproductive as well as developmental toxicity. The IARC estimated its toxicity of causing cancer in 1994 and ranked acrylamide as potential carcinogenicity. So the pollution of food caused by acrylamide has aroused worldwide attention and many governments have started the investigation of acrylamide in food. Model system studies have shown that acrylamide is formed during the Maillard reaction of reducing sugars with asparagine, when starch-rich food is processed at high temperature. Many health care teas are rich in starch content and they need to be processed at high temperature to make it more fit to be used. Whether or not acrylamide will be produced during the heating process is a question that attracts our research interest. In this work, acrylamide content in cassia seed tea and barley tea was detected by using SPE-high performance liquid chromatographic method, a new pre-column derivatization-high performance liquid chromatographic method was also investigated for the determination of acrylamide. This thesis consists of four chapters:Chapter one:The formation mechanism, toxicity study, detection methods, influencing factors and suppression method of acrylamide were summarized.Chapter two:An SPE-HPLC method was established for the determination of acrylamide in cassia seed tea. Cassia seed tea was ground and extracted with water and then centrifuged at the speed of 10000 r/min for 20 min, and 1 mL of supernatant was passed through OASIS HLB Cartridge and Bond Elut LRC-PRS Cartridge and eluted with 5% methanol-water solution, acrylamide in eluate was determined by HPLC. The acrylamide contents in cassia seed tea of different producing area and date were compared.Chapter three:The content of acrylamide in barley tea was detected by high performance liquid chromatography, and the acrylamide contents in barley tea of different producing area and date were compared.Chapter four:A new pre-column derivatization-high performance liquid chromatographic method was investigated for the determination of acrylamide, which included the preparation of derivative reagent and the optimization of derivatization conditions. Good separation was achieved between derivative reagent and the derivative product by high-performance liquid chromatography.
