High performance liquid chromatography (HPLC) has a dominant position in the field of drug analysis. Due to the advantages of good permeability and easy preparation, the monolithic columns recently show more and more important application in the pharmaceutical analysis. This work summarized the development of the polymeric monolithic column and its related applications.Preparations of the polymer monolithic column was investigated using glycidyl methacrylate as a monomer, ethylene glycol dimethylacrylate (EDMA) as a cross link agent, and AIBN as a initiator under a porogenic solvent of 1-hexadecanol. Moreover, the obtained column was further modified by temperature-sensitive poly N-isopropylacrylamide and successfully applied to the separation of immunoglobulin from human plasma.Moreover, another novel monolithic column synthesized by vinyl ester resin as a monomer, ethylene glycol dimethyl acrylate (EDMA) as a cross link agent, AIBN as an initiator, and 1-hexadecanol used as a porogenic solvent was introduced. The obtained monolithic column was further modified by poly N-isopropylacrylamide and successfully applied to the separation of lysozyme from egg white. Therefore, this kind of monolithic column is suitable using as stationary phase for HPLC.
