CD82molecμLes as the member of four transmembrane super family (transmembrane4super font-family, TM4SF) can express in many organizations, CD82can be widely involvedin cell growth, adhesion, apoptosis, and other life activities, highly conserved in evolution.Lamprey is the most primitive jawed vertebrates, is the bridge of communication ofinvertebrates and vertebrates, Lamprey is the bridge of communication of invertebrates andvertebrates, also is Model organisms in pursuing the development of the origins and evolutionof vertebrate immune. Lamprey is a pursue the development of the origins and evolution ofvertebrate immune model organisms. The main study concentrate at higher vertebrates CD82,but it is not clear that CD82exert what function and regμLating mechanism, because researchof some relatively low jawless vertebrates is less.From Japanese lamprey fat body cDNA library found a EST sequence that CD82molecμLes have high homology with mammalian CD82.Using primer5.0software designedprimers, gene cloning methods got CD82gene coding sequence. Japanese lamprey openreading frame of CD82gene is801bp, encoding266amino acids. Bioinformatics analysisshowed that CD82has four transmembrane domain structures (TM1-TM4), TM1betweenTM2form a small extracellμLar loop EC1, TM4and TM3extracellμLar form a large part ofEC2, and EC2has three potential N glycosylation sites. Using blast, Bioedit select differentspecies of CD82for sequence alignment, in addition, also predict CD82advanced structure,the distribution of antigen epitope and conservative motif. Lampreys CD82is the closestrelative with fish, completely following the order of creature from low to the evolution ofhigher.Using semi-quantitative RT-PCR method to detect genes CD82lampreys widelyexpressed in different organizations, the highest of abundance in leukocyte and kidney, in thefat body, gills, and other organizations also have different levels of expression, expression ofminimum in the heart. Using microbial immune lampreys, CD82mRNA expression levelsshow a trend of rise significantly in heart and leukocyte.This resμLts showed that CD82inlampreys immune system plays an important role in signal transduction, for the betterstudying lampreys laid a theoretical foundation and the evolution of the immune response.
