In recent years, Southern rice black-streaked dwarf virus(SRBSDV) causes huge damage on rice production in South China, and brings about much economic losses. Applying insecticide and traditional antiviral drugs were main means in rice field to guarantee rice normal growth. At present, there is no an effective method for screening anti-SRBSDV drugs, the key to inhibit SRBSDV is how to screen anti-SRBSDV drugs, so developing an anti-SRBSDV compounds screening model base on protein targets is significant in controlling SRBSDV.Prokaryotic expression method was used to acquire the P9-1 and regarded P9-1 as the target protein, researched the interaction of P9-1 protein with antivirals by using ultraviolet spectroscopy, fluorescence spectroscopy, isothermal titration calorimetry(ITC) and microscale thermophoresis(MST). Thus, the affinity between antiviral drugs and SRBSDV P9-1 was selected to evaluate the antiviral activity.From the series of experiments and analysis, dufulin, ningnanmycin, lentinan, amino oligosaccharin can bind with P9-1 by noncoleven interaction, their association constant were 1.51×105, 5.60×104, 4.78×104 and 5.60×103, respectively. The affinity between dufulin and SRBSDV P9-1 is the highest. Dufulin derivatives GUJHZX-013 and GUJHZX-025 also have stronger affinity with SRBSDV P9-1, their association constant were 9.12×104 and 1.41×105, respectively. But GUJHZX-005, GUJHZX-031, GUJHZX-035, GUJHZX-041 can’t bind with SRBSDV P9-1.
