In vitro micropropagation is an important biotechnology and is widely used in rapid vegetative propagation of plant, however it cost is so high that I can hardly use it in commercial purpose at large scale. AIoe.vera L.var.chinensis(HaW.) Berg. is used as my trial material, we research a new rooting method of plantlet cultured in vitro, with which the cost of plantlet cultured in vitro can be reduced significantly. We try to establish a micropropagation differentiation system which can be fit to root in gas.and get opium multiplication medium and synchronism medium for strong sprout; Research four factors such as sugar concentration, time of immersing, rooting reagent concentration, time of synchronism, which as main factors can influence one culture method significantly, get one culture conditions what the rooting rate can reach over 95%: synchronism time for strong sprout 27?0 day, sugar concentration 7.4O/o 8.2%, time of immersing I .79 2.85 hour, IBA concentration 59.46?2.35mgIL; Also explore the possibility of open operation rooting in gas, and select fungicides, MIC value of bacteriostat Baristim Derision,TOPsin M and DconiL278 is determined: SOOmg/L, 600mglL, 8OmglL, and research it effect on rooting culture, establish a method which open operation rooting in gas on condition mixing Baristim Derision and treatment fluid; By researching transplant of rooting in gas plantlet establish consummate rooting in gas system of plantlet cultured in vitro of Aloe. Root origination and comparative anatomical structure were observed, we testify that there are mainly differences between the method rooting in gas and common method, and explain the reasons why different culture method can get different rooting rate and survival rate..After checking cost value, we can know that one method cost value of rooting priod just is the last method cost value 25.5%,Cost value of rooting in gas p]antlet cultured in vitro reduce at 38.4%.So we can say that the rooting method is economical ,effective, and rapid.
