Sweet cherry is a kind of berries, which has an abundant nutritional value and a high economic benefit. However, the seedling production cannot meet the demand because of several problems of the traditional propagation. To provide a reference for the seedling production of sweet cherry ’Mingzhu’, the present study used the plant nodes as experimental materials to investigate several factors affecting proliferation and rooting of ’Mingzhu’ in vitro.BA concentration crucially affected shoot proliferation of ’Mingzhu’ in vitro, 0.5 mg·L-1 BA is favorable. Multiple shoot number and shoot growth were promoted when MS medium supplemented with the combination of 0.5 mg·L-1 BA and 0.1 mg·L-1 IBA. The culture vessel used the cap without ventilation was more suitable for proliferation of ’Mingzhu’ in vitro.In addition, raft bioreactor culture can produce mass and healthy multiple shoots, this method was better than traditional tissue culture for ’Mingzhu’ proliferation.The present study investigated the effect of MS medium strengths on rooting and plantlet growth in vitro, the normal strength of MS medium was optimal when compared with media lower than MS strengths. For rooting, NAA was more beneficial than IBA,0.5 mg·L-1 NAA was optimal. During transplantation, the substrate of humus soil, perlite, and vermiculite (1:1:1) was suitable for acclimatization of ’Minazhu’ nlantlets.
