Sensitive ELISA methods were established to detect somatostatin and HBsAg in JiSheng-I(JS-I) vaccine . The parameters reflected the proliferation of JS-I in avian embryo. The results showed the ratio of SS and HBsAg of expression product of JS-I was about 200-250:1 which was fundamentally accorded with result with RIA. The genetic engineering expression product was extracted with the osmosis shock method. With this produce the technique of ELISA was established that test anti-SS in sera from animals immunized with iS-I vaccine. Besides, another ELISA was set up to examine HBsAg antibody. The sera from animals immunized with JS-I vaccine were examined. The results showed the SS antibody and HBsAg antibody could be detected by our methods. Thought the experiments, the most suitable dose of JS-I vaccine to swine was determined: 1 O7PFU per swine. Two and ten weeks after immunity, weight gains of 1O7PFU group was 16.85% and 10.1% greater than those injected with saline (P<0.05). The groups immunized with those doses deviated from 10扨FU per swine were unsuitable. Additionally, the weight gain of swine was affected by the factors of management and disease. With pET-32 fusion protein system in E.coli somatostatin genetic engineering vaccine had been developed ,which could be used to immunize animal many times. There were two products expressed: 55-N/S fusion protein and SS-NIX fusion protein. Here they were respectively mixed with Freundincomplete adjuvant in proportion to 1:1 to immunize BALB/C mice. The result showed: SS-N/S and SS-N/X vaccines (i.e. JiSheng N/X and N/S vaccine) could obviously improve the growth rate of mice compared with the control. There were, however, a marked difference in response between sexes; The males failed to respond to the treatment, while the treated females grew faster than the saline controls .The weight-gain of the females immunized with 55-N/S or 55-NiX vaccine was 30.54% or 36.88% greater than those injected with saline. With the special coated technology of antigen, the new and sensitive test method was set up to examine anti-SS in sera from animals immunized with JiSheng N/X and N/S vaccine. The experiment showed the method was special. The anti-SS test proved immunized animals was induced to produce SS antibody, and SS antibody could last 3-4 months.
