In order to solve the problem of identification and classification of banana, and provide a scientific standard, based on this AFLP optimum system and program, molecular system , genetic diversity, genetic relationship and identification of the 35 germsplasm resources of banana from Guang Zhou Banana Germplasm plot were studied from Feb. to Dec in 2000. The main results from AFLP fingerprintings and cluster analysis indicated: High molecule weight DNA can be obtained from different organ of banana (spire, climax leaf, new root). The spire of banana is optimum for making template DNA in studies on identification and classification of Dwarf Cavendish base on AFLP fingerprintings. High molecule weight DNA and AFLP fingerprintings can be obtained by using the improved methods of SDS and CTAB. The method of improved CTAB and SDS(extracted with CH3C1 2 and 3 times) are better and DNA obtained by above methods has better quality, higher yield and clearer finger map . (3)The polymorphism bands can be obtained from three Primer pairs(EcoR I ACC+Mse I CAT, EcoR I ACC+Mse I CAG, EcoR I AAC+Mse I CAT), and EcoR I ACC+Mse I CAT is best in studies of banana genetic diversification, variety identification, finger map building, identification and classification with AFLP molecule markers. Different bands of EcoR I ACC+Mse I CAT AFLPfingerprinting of 35 materials strains of Dwarf Cavendish can be clearly identified. The number of amplified band swith AFLP primer pairs EcoR I ACC+Mse I CAT,and EcoR I ACC+Mse I CAG on 35 Dwarf Cavendish materials is 107 bands, the rate of polymorphism band is 78.5%, and the identified rate is 100%. Based on the cluster analysis of analyzed AFLP fingerprintings, 35 materials of Dwarf Cavendish from Guang Zhou Banana Germplasm plot can be divided into 7 groups when coefficients. Dice Similar measure is 0.88. 〥ice Similar coefficient of samples (B55,B59,B61) is 1.0. That is (to say), their genetic diversity is uniform, (7)35 Dwarf Cavendish materials have their own characteristic band in their AFLP finger map ,the characteristic bands of B58,B51,B24,B29 are obvious.There results play an important role in solving effectively on this problem the identification and classification of banana.Msc.
