AbstractSix Blood protein loci and LDH isoZyme polymorphism of Xinjiangdonk, Liangchou and Yunnan donk were detected by simple randomsampling. Polymorphic loci allele frequencies were Calculated and theirparameters of reliability and precision were given.Polymorphisms of blood protein and LDH isoZyme from Xinjiang donk,Liangzhou and Yunnan donk were detected by using PolyacrgIamid geleletrophoresis (PAGE). The results showed that Alb,Tf and AkP loci ofXinjiang and Liangchou donk were polymorphic. The pohymorphisms of Es,Pre and Poa loci were not detected out. But fOur loci of Yunnan donk wereploymorphic-lb, Tf, AkP and Pre. Tf Ioci of Xinjiang donk wascontrolled by 5 multip1e a1leles, but 4 alleles in Liangchou and Yunnan donk,There had no TP allele in Liangzhou donk and no Tf in Yunnan donk. Therest loci were all dominated by 2 alleIes. Five LDH isoZyme bands wereobtained in Yunnan donk, four bands in Liangzhou and Xinjiang donk. Buttheir character was LDH3>LDH2>LDH1 >LDH4>LDH5.Genetic variations of three populations were estimated by using averageheteroZygosity and efficient alle1es. The results showed that averageheteroZygosity and efficient alleIes were higher in Liangzhou and Xinjiangdonk. (H xinjiang donk=0.6l4, E xinjiang donk=2.96, and H liangzhoudonk=0.60l, E Liangchou donk=2.85). This showed that Xinjiang andLiangzhou donk seldom had artificial selection and their genetic diversitywere better.The minimum distance system cluster maP of gene frequencies at 2 bloodprotein ploymorphic loci in 8 donk breeds showed that the 8 breeds werecIassified as two grouPs. One was composed of Xinjiang, Liangchou andYunnan donk, and the distance was the shortest between Xinjiang donk andLiangzhou donk.The other group contained Guanzhong donk, Jinnan donk,Qingyang donk, Dezhou donk and Jiami donk. They were clustered a groupgradually, Which was accord with breed history eco-geograPhic distributionand historical materials. The results proved that phylogeny taxonomy usingblood protein polymorphism is scientific. The shortest distance clusteranalysis was adopted to analyse the LDH isoZyme pertentage. The clusteredresult accorded with that of blood protein po1ymorphism,eco-climate andbody index,Which testified phylogeny taxonomy was also accessible withLDH isoZyme.According to the results of blood protein polymorphism, LDH isoZymepercentage and eco-climate cluster analysis, Xinjiang donk firstly spread toLiangzhou, Ganshu province (Today's Wuwei) and formed Liangzhou donk.Then some of them sPread to Ningxia, North of shaanxi, Shanxi, Innermogonolia, Liaoning and formed equivalent breeds. Some reached toGuanzhong pIain across Ganshu province and lastly formed Guanzhong donk.These donk of Guanzhong district gradually distributed to middle and eastemdistrict,formed larger donk.
