Preventing disease effective is an important application in plant protection and plant molecular physiology. On the command of lasting agriculture development, the promising ways are: inducing the plant acquired the Systemic Acquired Resistance(SAR) by elicitor and transformation the special gene into the plant. Because of lacking the resistance species against Verticillium dahliae Kleb in gossypium, we screen out several elicitors induced xinjiang cotton resistance, and compare the resistance in cotton seeding and cotton callus. Using rare elements,COO-etc, we induced two species : Xinluzao7 and zhongmiansuo24 at the 3 or 4 foliages time in cotton. According to test result, PAL and Chitins(CHI) are correlative substance against Verticillium dahliae Kleb in lab, and select 4 feasible elicitors : Si,SA,Fe++,La+. After using single elicitor and mixture induce cotton seeding and tissue, we detected the resistant substance. Same detections do in trans chitins gene callus. The data show: the expression of resistant substance against Verticillium in cotton seeding has remarkable increased, and the I dropped from 77.3 to 44.3, and the resistance of xinluzao7 elevated to T level. The effect inducing by mixture elicitors was not the same as the single elicitor. After inducing, the expression of PAL and CHI has the consistency in cotton seeding and tissue. The total expression quantity in cotton seeding is larger than it's in cotton callus. Continual cultured two cycles, the level of resistant substance in inducing tissue come back to CK's in cotton tissue. In trans chitins gene callus, only CHI has obviously expressed and its quantity equal to resistant species zhongmiansuo24. The change of SA, which is possible signal in SAR, can detect in inducing cotton but not detect in trans chitins gene callus. The experiments provide that the elicitor can increase the resistance against Verticillium both in cotton seeding and callus; SA is one of signal elements when acquired systemic resistance in cotton; they are different resistant routes in plant through trans-gene and SAR.
