Inerleukin-2 (IL-2) was used as adjuvant to BOG and guinea pigs were used as the experimental animals in the study. The experiments were conducted to compare the immune modulation effects and histological lesions of the IL~2 adjuvant to Mycobacterium bovis BCG vaccine according to the levels of humoral immunity .cellular immunity and histopathology in immune organs.All guinea pigs were divided into three groups which were the BCG group , the expriment group, the cnntrol group. 0.1ml of M. Bovis BCG was subcutaneously into the guinea pigs in BCG and exprimental groups , respectitively and 0. 1ml of salt solution in control group. The experimental group were muscularyly injected with IL-2 at intervals. The results were as follows:(1) The results of cellular imnune assay (MTT)The sixth week after the vaccination , peripheral blood lymphocytes isolated were tested by IfTT . The result showed that the nonspecific OD value and the specific (X) value of guinea pigs of IL-2 group was significantly higher thanthose of BCG and sham injection group. At the sixteenth week after the vaccination , the second IL-2 was injected to the IL-2+BCG group . The OD values of IL-2+BCG group was higher than those BCG group and sham injection group again. The result suggest that IL-2 can enhance the nonspecific cellular and the specific cellular inmunity of the guinea pigs induced by BCG.(2) The results of humoral immune assay (ELASE) The second week afeter vaccination , the guinea pigs were detected by EILSA . The result showed that the OD value of guinea pigs of IL-2+BCG group had no sigificant difference with BCG group . It suggest that IL-2 can't effectively enhance the humoral immunity of guinea pig induced by BCG. (3) Results of hisopathological lesions of immune organs The lymphofollicular proliferation in T lymphocyte regions of spleen and lymphnodes of IL-2+BCG group were significantly higher than that of BCG and sham injection groups. This result further indicate that IL-2 can effectively enhance the cellular immunity of guniea pig induced by BCG.
