The objective of this study is to investigate the progesterone effects on the immune condition of mice uterus. During the study, medicines and chemical reagents were given at 7:00 p.m.in day 5 or day 9 of gestation.CDmifepristone was given to block the biding of progesterone to its recepters;(2)salin was given;(3)progesterone was given;(4)the Chinese herbal medicine prescription composed of Radix scutellariae and Rhizoma atractylodis was given at day3,day 4,day5,then mifeprestone was given at day 5;(5)progesterone was given at 8h after the mifeprestone had been given. Three mice were killed at 4h,8h,12h,16h or 24h after 7:00 p.m. at day 5 or day 9 of gestation every group. Uterine NK cells and CD8+ T lymphocyte were analyzed by immunohistochemistry and IFN- Y and TNF- a contents were analyzed by Enzyme Linked Immunosorbent Assay (ELISA), respectively. After the mifeprestone had been given at day 5 of gestation, IFN- Y content increased (P<0.01),and the number of NK cells increased significantly ;at day 9 of gestation , IFN- Y and TNF- a content increased(P<0.05) respectively, the number of NK cells increased significantly (PO.05), but less CD8+ T lymphocytes were counted(P<0.05).Progesterone was given at day 5 of gestation, IFN- Y content decreased significantly (PO.01), more NK cells were counted and more CD8+ T lymphocytes were counted (PO.01) in the uterine Muscle; while at day 9 of gestation, IFN- Y content increased significantly(P<0.05),less NK cells were counted(P<0.01), and less CD8+T lymphocytes were counted (P<0.01) in the endometria and muscle of the uterus .In the herb prevention group, at day 5 IFN-Y increased, less CD8+ T cells were counted in endometria and muscle of the uterus (PO.01 ) ; At day 9, IFN-Y content and the number of NK cells increased significantly(PO.01),less CD8+ T lymphocytes were counted (PO.01), but IFN-Y and TNF- a contents had been intent to decrease at 4h. the result therefore indicate that progesterone of the Physiologic content inhibits the extreme increase of IFN- Y andTNF- a contents in the uterus .That mifepristone was given to block the immune condition develops to Thl-type response .So that IFN-y content increased in the uterus, NK cells gather and infiltrate, those induce the embryo loss. And the effect on the abortion at day 9 of gestation is more obvious than that at day 5.In the progress of normal pregnancy, the NK cells, IFN- v and TNF- a in the endometria benefit to the embedding of embryo and the maintence of pregnancy, progesterone may increase the number of NK cells and CD8+T lymphocytes at day 5 of gestation. Though progesterone and the Chinese herbal medicine prescription composed of Radix scutellariae and Rhizoma atractylodis couldn't entirely prevent the mifeprestone-induced abortion.that have a certain antiabortive effect.
