Studies On Storage Protein In Mythimna Separata And Effect Of Celangulin V On Storage Protein

Storage protein is a kind of specific haemolymph protein and plays many roles in the insect whole lives.The studies on the kinds of storage protein,molecular weight,subunit molecular weight,compositions and contents of amino acid of storage protein,and the sublethal effects caused by celangulin V on the contents of storage protein and the mounts of laid eggs by the adults had been carried out by means of PAGE,SDS-PAGE,Western blot and amino acid analyzer.The main results are as follows:The storage protein was detected in 1 day old last instar larva of Mythimna separata.The concentration of storage protein in the haemolymph increased with day old elongated and reached to peak values in the last day old.The storage protein was taken up by the fat body,so that it decreased in the haemolymph.There were two kinds of storage protein termed SP-1 and SP-3 in the pupal.Only SP-3 was present in the adult stage.SP-3 termed as persistant storage protein appeared in the fat body of pupal,which could be expressed until the adult stage.SP-1 was a hexamer,which was composed of two subunits ,which were calculated to be 94kDa and lOOkDa respectively.SP-3 was composed of subunit which in size of 94kDa.These two hexamer storage proteins in native molecule weights are 550kDa.The amino acid compositions of SP-1 was similar to SL-1 of Spodoptera litura and having c. 8.16% aromatic amino acids(phe and tyr),SP-3 with composition differed from which of SL-1,having c.3.06% methionine.The contents of the storage protein in 1 day old,2 day old,3 day old,4 day old,5 day old of the treated last instar larva were markedly lower than the control larva Mythimna separata .After 6 day old,no difference was tested in contents of the storage protein and the mounts of laid eggs by female adults between the control and treated by celangulin V.
