The most important thing in the colored cotton production is the poor quality of colored cotton fiber, Far from meeting the textile request and people' s wanted in daily life. Face this situation, problem that should be first solved is to improve the intensity of fiber of colored cotton. People have studied the physiology mechanism and biochemical mechanism of cotton fiber form. Carring on extensive research to cotton fiber relevant genes of development, have got a series of cotton fiber development relevant genes and some promoters be of peculiar in cotton fiber.In this study,we choose the prp , that is related to accumulation of secondary wall material of cotton fiber cell, and E6 promoter, the promoter is of peculiar in cotton fiber cell, to constructer the plant expressed vector. Acqurie E6 promoter fragment from pBI121 plasmid vector, and coloned it into pMON10098 vector that have cutted down P-35S promoter fragment. Positive clones were analyzed with restriction enzyme digestions. The enzyme digestions results show the E6 promoter fragment have coloned into pMON10098 vector, and obtained pMONE6 vector.With a set of primers designed according to the sequence of prp , the 3' and 5' ends of the primers designed new enzyme digestions sites, the prp fragment were abtained by Polymerase chain reaction. Then coloned it into pMONEG vector,. Positive clones analyzed by the PCR and restriction enzyme digestions analyzed, the results show prp fragement have coloned into pMONE6 vector. The vector can be used intransgenic meidiated by particle bombardment and Agrobacterium.According to reports, compared several transgenic methods, selected particle bombardment mediated transgenic methods, through experiment try to find out and set up shoot apex regeneration system. Choose sterile seedings on 4-5 days age, the shoot apex regenerated rate is most high. The shoot apex regeneration is can be find in MB culture medium without hormone. Graft raise can improve survival rate of seedlings , the shoot apex survival rate is up to 80%. By the graft raise and the transgenic method of particle bombardment combined,we set up a set of convenient and swift transgenic system of cotton. Through this system, prp have transform into xincaimian3 by particle bombardment mediated method., The results of PCR analyzed shows the prp fragment have been transferred into chromosome of xincaimian3 already.
