Isolation And Identification Of Porcine Pseudorabies Virus HN Strain And Cloning And Sequence Analysis Of The TK Gene Of The Strain

One virus strain was isolated from the brain and viseceral organs of diseased piglet from a pig farm In Zhengzhou.The virus induced typical cytopathogenic effect(CPE) after innoculated in BHK cell line. Affected cell melted, formed giant cell. The virus could be neutralized by the positive serum to pescudarabies virus (PRV). With the method of Reed-Muench' s, we performed neutralizing test.The titer of the virus strain is 1: 107, and the titer Of PRV E A strain is 1:209. Two adult rabbit were injected with the virus and the typical pseudorabies clinical syndrome could be observed. All of the evidence indicated that the isolated virus was PRV.A pair of primers for PRV isolated strain was designed according to published PRV TK gene sequence. With using of PCR technique, TK gene of the PRV strain was completely expanded. The whole TK gene was cloned into pGEM-T-easy vector and the recombinant plasmid was idendif ied by enzyme digestion.
