Corn endosperm ribsome inactivating protein(CRIP9) gene is transferred into sugar-beet(Beta vulgaris) by means of plant tissue culture and plant genetic engineering.The purpose is to obtain a new variety(strain) of sugarbeet(Beta vulgaris) that has resistance of disease. At present .certain achievements have been obtained as following: (1) The regenerative system of high frequency using the explant of petiole has been successfully developed by switching culture medium. In the two culture media,the frequency are 50% and 61% respectively. The differentiation culture medium is MS basic medium supplemented with 0.75mg/L 6-BA or plus 1.0mg/L 6-BA and 0.3mg/L NAA, root induction medium contains 1/2MS supplemented with NAA1.0mg/L;(2)The steady system of genetic transformation has been developed for sugarbeet(Beta vulgaris),using Agrobacterium tumefaciens and obtains resistant shoot.All these have established the base for acquiring transformed sugarbeet(Beta vulgaris) and cultivating the new variety (strain) of sugarbeet(Beta vulgaris) that has resistance of disease.Co-cultivation medium is differentiation culture medium supplemented with Rif 40mg/L and SP 100mg/L. Selective differentiation culture medium contains differentiation culture medium ,Kan100mg/L and Ce 500mg/L.
