cRIP genes were transferred into Sugarbeet via Agrobacterium using petioles as explants. The factors that affected transformation frequency were investigated ,such as preculture time, concentration of Kanamycin(Kan), concentration of Cefazolin sodium(Cef), infection time and infection concentration of Agrobacterium ,co-culture time, concentration of AS etc. When preculture time was 2 days ,infection concentration was OD600=0.3-0.4 and infection time was 8-10 minutes and co-culture time was 2-3 days ,high transformation frequency can be obtained. At the same time , 200 ug/L AS was added into co-culture medium and 100mg/L Kan and 500mg/L Cef were added into the seclective medium. The highest frequency was 36.0%. A stable and efficient Agrobacterium mediated transformation system has been established which offers a base for high yield,high quality and broad resistance of disease Sugarbeet variety.
