| Chicken interferon gamma(ChIFN- γ) is an important cytokine. The research was to detect the effect of rChIFN-γ on intestine immune response of broilers immunized with E.tenella oocysts through immune technique.Recombinant ChIFN- γ was expressed in vitro. After being purified and renatured, it was tested for activity with CEF-VSV. The result showed that the anti-viral activity was 210U/ml. This research established a foundation for large-scale production of ChIFN-γ.In the study of the effects of rChlFN- Y on the vaccine immunity, the results showed that rChlFN- γ could improve height of intestinal villus.crypt depth and thickness of brush border of broilers immunized with E.tenella oocysts through H.E and Pas technique. At the age of 21, all the data of broilers immunized with rChlFN- Y were significantly higher than that of immunized control broilers. The data of broilers immunized with rChlFN-γ were 3.25mm, 115.8um and 21.1 urn separately,whileas broilers without rChlFN- Y treatment were 2.57mm,95.9um and 19.1 urn.At the same time, the number of IEL and GC of broilers immunized with rChlFN-γ were significantly higher than that of the immunized control broilers, which were 9.8 versus 8.2 and 26.2 versus 24.9. This means that rChlFN- Y could increase the proliferation and activation of IEL and GC.rChlFN-γ can elevate mast cell number of duodenum, caecum , jejunum and ileum. At the age of 21, the results of broilers immunized with rChlFN- Y were significantly higher than that of the immunized control broilers, which were 715 versus 693,86.4 versus 78.0,209.6 versus 172.0,and 96 versus 81.6.The results of slgA level showed that rChIFN-γ can improve sIgA level od broilers' intestine.The results of our research demonstrated that rChlFN- Y can increase the proliferation and activation of IEL, GC and MC.inprove slgA level, enhance the intestinal mucosal immunity. rChIFN-γ could be an excellent candidate as potential adjuvant for coccodia immunity.
