Tomato is perennial herbaceous plants which belongs to Solanaceae Solanum,coming of Peru in South America, it is one important kind of popular vegetable in the world. The objectives of this study are to develop superior cultivars, and to effectively conserve and utilize the superior genetic resources, to develop methods of identification at molecular level for new variety registration of the species. In this study, analysis of genetic diversity, study on genetic evolution and fingerprinting of sixty-two elite tomato cultivers were carried out with AFLP markers, and carried out clustering analysis of these sixty-two elite tomato cultivers. The main results as follows:1. Amplification testThe primer combinations P+3/M+3 can amplify a proper number of bands and the AFLP fingerprinting was very clear after the sliver -staining.2. Primers selectionP01/M02 primer combination was used in preamplification and P+3/M+3 primer combination was used in amplification. Sixty-five pairs of optimum primer were selected from two hundrand and seventy pairs of primer. The high percentage of polymorphism and abundant bands were obtained by using these sixty-fve pairs of primer in AFLP amplification. In every primer, it can get two to four polymorphism segments.3. the construction of tomato AFLP fingerprinting testingTo carry out the AFLP amplification with thirty-two pairs of primer that selected from the sixty-five pairs of screened optimum primer, and to use five of them, whose polymorphism information contents are richer than others, basing on the data that got from amplification results, the AFLP fingerprinting of the sixty-two tomato cultivars was established.4. The genetic diversity analysisBasing on the data statuted in the amplification test,the genetics analysis for all the seleced sixty-two tomato cultivars were carried out,and the clustering analysis was established.
