Genetic diversity of eight rabbit lines was evaluated on the basis of 6 microsatellite markers .The PCR amplified products of microsatellites were detected by denatured polyacry lamide gel electrophoresis .Allele frequency .polymorphism information content , gene heterozygosity for six microsatellite in eight rabbit breeds loci were calculated , and genetic distances between these breeds were estimated . And then , cluster analysis based on those calculated genetic distance lines was performed for the eight rabbit line.The results showed that every microsatellite loci analyzed were polymorphic in those rabbit lines with an average number of alleles per locus and the average polymorphism information content (PIC) equal to!3 and 0. 5300 respectively. Rates of heterozygosity in eight line are all high , and that is the highest in New England Cottontail(0.6818) .which suggested that those rabbit breeds had abundant genetic diversity and potential of selection. The result of genetic distance indicated that there were the close genetic relationship among those rabbit line, the genetic difference between California rabbit .Belgium, and Tianfu black rabbit is same to their relationship with Harbin white rabbit, that is consistent with breeding history. In regard to the results of UPGMA cluster analysis according to genetic distances, itcouldn' t reflect the known relationships and genetic variations between any two populations. Microsatellite markers Satl2^ SOL28 all presented null alleles and SOLOS acted as duplicated loci obviously during PCR amplification in some individual DNA samples. To reach an accurate result, all the markers to be used in a study mustbe selected properly, and the genetic variations from much more loci in genome will be needed to get enough information in similar study in the future. These results could provide, basic molecular data for research on genetic diversity and QTL of rabbit.
