In this study, ten microsatellite loci were used in this experiment to analyze the genetic diversity of 6 rabbit populations including 3 native rabbit breeds and 3 commercial lines. The thesis discussed the genetic variation within the population and among the populations which can provide the scientific theory of protecting and using of rabbit resource.In the experiment, the PCR amplified products of microsatellites were detected by denatured polyacrylamide gel electrophoresis (PAGE), and the genotype was judged from the electrophoretograms. Allele frequency, polymorphism information content (PIC) and heterozygosity value (H) often microsatellite loci in the 6 rabbit populations were calculated, and genetic distances between these populations were estimated. Then cluster analysis based on those calculated genetic distance was performed for the 6 rabbit populations.The result shows that every microsatellite loci analyzed are polymorphic in the 6 rabbit populations with an average number of 4.8 alleles per locus, the polymorphism information content and heterozygosity value are between 0.6429~0.7800 and 0.4130~0.9941 respectively, which can acquire the conclusion that these microsatellite loci can provide the most information of genetic diversity and can be used firther. All the heterozygosity values of 6 rabbit populations are high which are from 0.8133 to 0.9035, and the value in Bulietaniya rabbit is the highest, which show that these rabbit populations have abundant genetic diversity and the potential to be selected. The result of Ds and Da genetic distance shows that the genetic distances between the 6 rabbit populations vary inapparently. According to Ds and Da genetic distance, we made the NJ and UPGMA cluster respectively. Comparing the 4 dendrograms, we can read that the results have little difference. In the two UPGMA dendrograms of Ds and Da, Bulietaniya, Wanzai and Sichuanbai rabbit are in close relationship,Yila, Yipulv and Fujianhuang rabbit are farther from the other rabbit populations. In the two NJ dendrograms of Ds and Da, Yipulv rabbit and Yila rabbit have both the fartherest relationship with the other populations which are consistent with the result in the UPGMA dendrograms. But the relationship of Fujianhuang rabbit with the other rabbit populations is different in the 4 dendrograms.
