The Study On The Telomerase Activities Of Erythrocytes,Lyph-ocytes,Serum And Spleen In The Development Of MD

Telomerase is a ribonucleoproteinase (RNPase) composed of telomerase RNA and telomerase protein. It can recognize and combine with the end of the telomere and synthesize telomere by reverse transcription, using its RNA component as template. The RNA subunit of telemere is important for its structure and catalyzing activitiy and acts as the template of telomere DNA synthesis. In most of the somatic cells and primary cells, the activity of the telomerase is very low and undetectable. However, the telomerase is activated in tumor cells, which is essential for cell division and overcoming the continuous loss of telomere sequence. MD is lymphoproliferative disease. It is caused by herpes virus of chicken and characterized by tumor formation in visceral organs. Recent studies have proved that the sequence of MDV is similar to telomere sequence of the host cells. Therefore we use modified TRAP assay to detect the changes in telemerase activities in the develop- ment and progress of MD tumors so as to probe into the pathogenesis of MD regarding to telomerase activities. The establishment of MD tumor model82 of Hailan Chicks of 1 day were divided into 2 groups randomly. 41 of the chicks in experimental group were inoculated intra-peritoneally with 0.5ml blood containing MD-1 virus and the other 4 of control with saline on the 1 day. From the 6th day, the chicks were put into death after taken blood from the hearts. The lymphocytes were isolated with separating medium and then erythrocytes and serum were collected. The spleen was taken out and stored in liquid nitrogen. The detection of the telomerase activity of MD tumor cellsThe telomerase activities of MD tumor cells in 1 cases were detected with modified TRAP and the results showed that the telomerase activities were high in MD tumor cells. 昑he detection of the telomerase activity of the blood from chicksThe telomerase activities of lyphocytes, erythrocytes and serum from 41 chicks with MD tumors were detected with modified TRAP. The results showed that the telomerase activities in lymphocytes and serum began to appear on the 8th day in the challenge group. The positive rate were 63.41% and 58.54% respectively. No telomerase activities were detected in the control. The detection of telomerase activities in MD tissuesThe telomerase activities in spleen of 41 chicks were determined with modified TRAP. The results showed that the telomerase activities in spleen began to appear on the 7th day in the challenge group. The positive rate were 43.90%. No telomerase activities were detected in the spleen of the control.On the basis of the experiments, we concluded that that the telomerase activities are correlated with the stages of MD tumors.
