Antifungal Protein of Gastrodia Elata(GAFP) gene was used to transform the three Xinjiang upland cotton system without disease resistance gene via the pollen tube pathway method. Then, through the mark gene positive experiment and inovulated disease bed, resistance to Verticillum Wilt of transgenic cotton was identified, and the totally disease resistant cotton plants were selected. After molecular test of Southern dot-blotting, the results showed that:1. Using the GAFP gene to transform the three Xinjiang upland cotton system without disease resistance, disease resistance materials whose DNA were integrated could be obtained.2. The technique of Southern dot-blotting could be used for testing transgenic generation whose DNA was supplied by GAFP gene . After the Southern dot-blotting molecular test of 25 identified plants, in two plants there occurred the hybrid signal which testified the integration of GAFP gene.3. Pollen tube pathway method is an efficient and a rapid means of disease resistance cotton breeding and its transform rate is about 1%. Using this method, we obtained two transgenic plant.The results of this experiment showed that using the pollen tube pathway method, the GAFP gene could be transformed into cotton system and new totally disease resistant cotton plants could be obtained.
