Genetic markers have been an absolutely necessarily research means since modern genetic has been established this century. STR locus complies Mendelian inheritance principles and distributes abroad and amplifies with PCR and polyacrylamide gel analyzed with sliver staining, so it is applied in molecular genetic science fields and developed startling. Individual identification and parentage tests are improved fast using microsatellite markers. The aims of the present study were:(1)to study the polymorphism distributions of six STR loci in dairy cattle and beef cattle;(2)to establish a nicety and shortcut method for and parentage tests in dairy cattle and beef cattle.1 The polymorphism distributions of STR loci in dairy cattle and beef cattleCatties for experiments were from two breeds, including 9 Holstein Friesian bull tissue samples and 49 Holstein Friesian cow blood samples and 103 beef cattle tissue samples. Extracted DNA from these samples and analyzed the polymorphism distributions of six STR loci in dairy cattle and beef cattle. The polymorphism distributions of six STR loci, BM2113, BM1862, BMc701, BM2934, TGLA122, BM720 were detected in cattle by Polymerase Chain Reaction multiplex gel electrophoresis followed by silver staining. Gene frequency (Pi) , power of discrimination (DP), heterozygosity (H), polymorphism information content (PIC) and probability of paternity exclusion (PE) were calculated. All loci meet Hardy-Weinberg equilibrium. DP, H and PIC of BM2113 locus, and PE of TGLA122 locus are the biggest among six STR loci in Holstein Friesian. Cumulate DP of six STR loci is 0.99997, Cumulate PE of six STR loci is 0.98827. DP, H, PIC and PE of BM1862 loci are the biggest among six STR loci in beef. Cumulate DP of six STR loci is 0.99999, Cumulate PE of six STR loci is 0.99578. The six STR loci have high genetic polymorphism and they could be used as linkage analysis, individual identification and paternity test in cattle.2 Application of STR Loci by Silver Staining And PCR Methods for Parentage Identification in Dairy Cattle and Beef cattleAmplifications of six STR loci such as BM2113, BM1862, BMc701, BM2934, TGLA 122, BM720 were performed by PCR (polymerase chain reaction) method. The products of amplification were separated in polyacrylamide gel and analyzed with sliver staining. The method used in the case could provide a power of paternity of approximately 99.993%. The results identifying a heifer's paternity with known maternity showed that the sliver staining was a validation method for identification parentage in dairy cattle.
