In the study we still used microsatellite marker to test the parentage of shack-Kee.7 polymorphic microsatellite DNA primers identified in pigeons were amplified in shack-Kee. By all of the loci amplify products, we calculated the gene heterozygosity , the polymorphism information content , effective number of alleles and excluding probability of paternity of each locus. The results were as follows:In 187 samples, we found 36 alleles of 7 polymorphic loci and there are the most alleles in the ClimD16 and ClimD01 loci. We calculated the allele frequencies of 6 polymorphic microsatellite loci, and then we calculated the gene heterozygosity(H), the polymorphism information content (PIC) , the effective number of alleles and their mean of each locus by the allele frequencies, the results as follow: The values of gene heterozygosity is 0.650-0.812, the mean is 0.758 .The values of polymorphism information content is 0.583 -0.782, the mean is 0.715 .The effective number of alleles is 2.8521-5.2526, the mean is 4.2903. All of these results showed this population had richer polymorphism and genetic diversity .The results provided a basis for the later study.We succeed in parentage test in the population of shack-Kee by the 7 microsatellite loci. ,and corrected the mistakes in the pedigree , accommodated the breeding in the factory. At the same time we calculated the excluding probability of paternity (PPE) of each locus and cumulative chance of exclusion(CCE), the value of PPE was 0.374-0.620 and CCE was 0.9956.We got the right sex in 48 individuals of sex-known gray wild pigeons and in 216 individuals of sex-unknown shack-Kee - with CHD and EE0.6 by the technology of PCR and agarose gel.1 The primer 2550F/2718R of CHD gene sequence were used to amplify the gene or sequence in the sex-known gray wild pigeons, the results showed that there were two bands in female and one band in male, According to the results we can successfully identify the sex of sex-unknown shack-Kee , and cloned the sequence of CHD-Z and CHD-W gene fragment in shack-Kee.2 The primer EE0.6F/ EE0.6R of EE0.6 gene sequence were used to amplify the gene or sequence in the sex-known gray wild pigeons, the results showed that there was one band in female and none band in male, According to the result we can successfully identify the sex of sex-unknown shack-Kee , and cloned the sequence of EE0.6 gene fragment in the female shack-Kee.3 We analysis the sex in the pigeon which had the same age and the factor which could concerned with growth in the birds, the result showed that the sex had significant association with Carcass Weigh, Body Weigh and Holo-Carcass Weight in the population of the pigeon. (P<0.01).It proved that the results of sex identification were right.
