The Genetic Diversity Of 12 Microsatellite Loci In Three Police Dog Breeds And The Application In Parentage Testing

The purpose of this study was:1) to analyze genetic diversity of the imported police dog and provide a theoretical basis for breeds conservation. 2) to identify the parentage by the multiplex PCR,and further empolder domestic canine genotype kit lay the groundwork for the establishment of the police dog DNA database;3 ) to discuss the police dog DNA database so that it can be used for establishing marker-assisted selection method for breeding. Experimental animals included 54 German Shepherd dog, 57 Labrador Retriever dog, 26 English Springer Spaniel.1. Study on genetic diversity of three dog breeds population by STRVariability of 12 canine microsatellite loci named PEZ1, PEZ3, PEZ6, PEZ8, PEZ10, PEZ13, PEZ15, PEZ16, PEZ19, FH2010, FH2079 and FH2611 in different chromosomes were examined among three pure breeds of police dog such as German Shepherds, Labradors, English Springer Spaniel. Polymorphism information content (PIC), index of genetic diversity (H), expected heterozygosity (h(e)), observed heterozygosity (h(0))and effective number of allele (ne) were evaluated..The results showed that markers PEZ3, PEZ6, PEZ8, PEZ10, PEZ13, PEZ15, PEZ16, FH2010 and FH2611 were high polymorphic; Marker PEZ19 was middle polymorphic in Labradors, while in other breeds of police dog was high polymophic; Marker FH2079 was middle polymorphic in English Springer Spaniel, while in other breeds of police dog was high polymorphic; Marker PEZ1 is low polymorphic in German Shepherds, while in other breeds of police dog was high polymophic; The evolutionary relationship among these closely related dog breeds were estimated by genentic distance measures developed for use with microsatellite loci. According to the paiwise genetic distances, German shepherds and English springer spaniel had longer diverse evolutionary histories than German shepherds and Labradors or Labradors and English Springer Spaniel.
