Chinese cabbage-pe-tsai (Brassica campestris L. ssp. pekinensis Olsson) is an important vegetable variety occurred originally in China. At home and abroad many researchers have put to use biotechnology in the field of Chinese cabbage-pe-tsai breeding work. At present pollen culture and isolated microspore culture are the most useful ones of biotechnology. This experiment is comparative analysis of the methods of pollen culture and isolated microspore culture. This paper studies some elements that effect on the culture and selects the most suitable method and parameter. On the other hand, this paper studies the practical efficiency of pollen culture and isolated microspore culture technique in the plant breeding work. This paper also comparatives the merits and demerits in order to provide some advises for improving the method and developing the efficiency for them to be used more widely in the plant breeding work. The main contents and results are as follows.(1) The difference of genotype is the key element affecting the producing of the pollen embryo of Chinese cabbage-pe-tsai. The difference of the efficiency of embryo inducement is very great. The result also indicts that the ability of embryo inducement is similar between high parent and FI(2) High temperature pretreatment can improve the efficiency of pollen embryo inducement greatly. The temperature of pretreatment having the best efficiency is 33℃ The time of pretreatment having the best efficiency is generally between 24 and 48 hrs.(3) Adding the content of organic matter can improve the efficiency the pollen embryo inducement of Chinese cabbage-pe-tsai. Reducing the content of organic matter is harmful for the inducement of the pollen embryo. The concentration of sucrose having the best efficiency in the medium of pollen culture is 10% and it is 13% in the medium of isolated microspore culture. The medium without hormone has the best efficiency for all.(4) The efficiency of regeneration of pollen plants with the pollen embryo inducement in pollen culture is higher greatly than that of isolated microspore culture. The amount of pollen plants regeneration from a bud in the isolated microspore culture is higher slightly than that in pollen culture.(5) The embryo inducement efficiency of isolated microspore culture is decuple higher than that of pollen culture. But the efficiency of regeneration of pollen plants using isolated microspore culture is lower greatly than that of pollen culture. The technique of the isolated microspore culture has more difficulties and need more costs than that of the pollen culture. So many key sectors of the technique of isolated microspore culture should be improved as the technique to be widely used in the factual research work.
