| Varieties bred from ber, Zizyhpus mauritiana Lam. (Taiwanqingzao) were known as a new economic fruit tree in tropic and sub-tropic zone of China, grafting is effectual ways for their propagation in orchard. The germination percentage of ber is very low, the grafting affinity is not good by native or on Zizyhpus spinosus Hu. (suanzao) rootstock, this would be overcome by using In vitro culture for their rapid propagation. The results were reported in this study.1.Study on Sterilization of In vitro culture in Zizyhpus mauritiana Lam. (Taiwanqingzao)Effects of sterilization on explants and growth on callus were studied In vitro culture of Zizyhpus mauritiana Lam. (Taiwanqingzao). 1/600 Carbendazim, 75% Alcohol, 0.1% Mercuric Chloride and Tween-20 combined to establish the Sterilization method. The explants's pollution rate was decreased by spraying 1/600 Carbendazim 5 days prior to the explants cutting. 75%Alcohol the explants for 30 seconds and 0.1% Mercuric Chloride with 3-5 drops of Tween-20 for 5 minutes for sterilizing, which not only decreased the explants pollution rate, but also enhanced the callus growth.2.Effect of Culture Conditions on In vitro culture of Zizyhpus mauritiana Lam. (Taiwanqingzao)Effect of temperature, illumination period, pH on In vitro culture of Zizyhpus mauritiana Lam. (Taiwanqingzao) was studied. The suitable temperature was between 30℃-32℃, the proper illumination period was about 24 hour and pH 6.0.3. In Vitro Culture of Zizyhpus mauritiana Lam. (Taiwanqingzao)The combination of different kinds and concentration of growth regulators were studiedaccording to callus production rate of explants, callus weight of explants in initial culturemedium and callus progression in subculture medium on basis of MS medium in Zizyhpus mauritiana Lam. (Taiwanqingzao). The proper initial medium: MS+2,4-D (0.5mg/L) +KT (0.1mg/L) (Young fruits); MS+2,4-D (1.0mg/L) +KT (0.1mg/L) (Budding stemstick)and subculture medium: MS+ZT (O.lmg/L) . |
