This paper researched the temporal spatial expression of Glucose Oxidase gene in trans GO cotton plants and H2O2 temporal spatial variation in cotton plants. It campared trans GO cotton varieties B99267-1 , B99261 with recipient control Xinluzao7 and conventional variety Zhongmiansuo 12 on the seedling disease resistance , boll disease resistance , Verticillium Wilt resistance and agronomic character of cotton plants.Glucose Oxidase activity in trans GO cotton plants varied in different stages and parts. At the stage of florescence, boll and harvest ,enzyme activity in the stage of florescence was the lowest and the stage of harvest was the highest.In all stages, enzyme activity in leaves of the top were usually lower than that of down.In cotton seedlings.enzyme activity in leaves were higher than roots and stems, and when being inoculated with pathogen of Verticillium dahiae, the GO activity in the seedlings increased, the roots and the stems of seedlings increased much significantly.GO activity positively correlated with GO protein and H2O2 level.Trans GO cotton varieties(B99267-l and B99261)were more resistant to Verticillium Wilt than the susceptible recipient control Xinluzao 7.The resistance capability of Verticillium Wilt in B99261(VIII) was lower than that in B99261(IV).And disease resistance of B99267-1 kept stable in all generations which was consistent with enzyme activity detection.When GO gene was transducted in cotton, the cotton plant became dwarf, the boll stems became longer, the number of bolls increased and the fiber attributes improved. In addition, trans GO cotton became more resistant to pathogen of Colletotrichum gossypii, Diplodia gossypina, Phytophthora boehmeriae, Rhizoctonia solani and Fusarium oxysporum.It was concluded that Glucose Oxidase gene was a wide-spectrum resistance gene.lt would have very important signification in Verticillium Wilt resistance breeding of cotton.
