Mycoplasmal pneumonia of sheep and goats was studied in rapid detection by multiplex polymerase chain reaction and prophylaxis-therapy in medicine. The aim is to find methods for the control of the disease in production.A multiplex polymerase chain reaction assay was optimized to stimultaneously detect Mycoplasma Ovipneumonia and Mycoplasma CapriPneumonia. Two sets of specific primer were designed according to nucleus acid sequence of 16S rRNA . It was showed that all samples contained Y98,HD stain and PG3 can be amplified by multiplex PCR using two set primers, yielding two specific bands of 466 bp (Y98,HD strain),273bp (PG3).But no specific band was amplified for Mycoplasma Pneumonia and other bacteria. As little as 10pg of Y98 and 1pg of PG3 DNA can be detected using gel electrophoresis in this multiplex PCR. 132 cases of clinical specimens were detected by using multiplex PCR, direct cultivation and specific serological tests.The results showed that multiplex PCR was more sensitive than cultivation. The detection of more positive by multiplex PCR than by specific serological test was not expected. But according to the results, it was showed that multiplex PCR was more sensitive of detecting clinical samples than serological test in early stage of its infection.The effects of these eight antibacterial drugs on M. myc. sub. Myc. PG3, MO HD strain and Y98 in vitro were showed by using the method of micro checkerboard titration . Their minimal inhibitory concentrations(MIC, ug/mL) were respectively: ciprofloxacin 0.223 0.00327 0.00223,ofloxacin 0.281 0.0104 0.140,danfloxacin 0.136 0.053 0.0140,eromycin0.0218 invalid invalid, roxihromycin 0.0327 invalid invalid, tylosin 0.0442 0.13 0.0390, taimulin 0.0517 0.091 0.0520, tetracycline 0.1950 0520 0.0520. The results showed that eromycin and roxihromycin were highly inhibitory on PG3.,and ciprofloxacin, danfloxacin and ofioxacin were highly inhibitory on Y98. The fractional inhibitory concentration(FIC) index of ofloxacin combined with eromycin or tetracycline on PG3 strain was 1 .The FIC index of tetracycline combined with ofloxacin or tylosin on Y98 was 0.5.The FIC index of tylosin combined with ofloxacin was 1.These results lay basis for the further development of new high efficacious and low-priced compound against mycoplasma pneumonia of sheep andgoats.The prophylactic and therapeutic efficacies of these drugs were observed in experimental infection with Y98 in sheep and PG3 in goats. The results concluded that danfloxacin arid taimulin was the first choice for the infection of Mycoplasma ovipneumonia, while eromycin and tylosin was the first choise for the infection of Mycoplasma capripneumonia. By muscular injection the prorapratic dose is 5 mg/kg body weight ,while the theraprutic dose is 10mg/kg body weight with a treatment period of 1-2 weeks.
