The tissue culture leaves of (P.simonii ×P.nigra)×R15 ACL has been studied. The development process of adventitious roots induced by 0.3mg .L-1 exogenous NAA was observed with paraffin mounting method. The result shows that the occurrence of adventitious roots esses three stages:1, induction stage;2. root primordium formatting stage; 3. adventitious roots differentiating and elongation stage.On the basis of tissue culture leaves adventitious roots occurrence different period appearance anatomy observing ,the content variation of endogenous hormones IAA, ZR, GA3 and ABA during rooting of leaves was measured with ELISA. The relationship between root generation and endogenous and exogenous hormones is discussed. It reveals that: the level of endogenous IAA ties up with adventitious roots generation. ZR doesn't influence adventitious roots generation observably, but plays an important role in consequently adventitious roots growth. Low content of GA3, high level of ABA and high ratio ABA/GA3 are useful to generation of adventitious roots.The changes of protein expression during different period of the tissue culture leaves of (P.simonii X P.nigra) X P.15 ACL development were studied with SDS-PAGE.The best method for extracting solube proteins from leaves was selected for polyacrylamide gel was established.The result showed that there exists two decrescendo protein bands with 76.3 X 103 and 64.1 X 103 during the livelong stage. It presents two new protein bands with 47.3 X 103 and 41.4 X 103 in adventitious roots differentiating and elongation stage.Two-dimensional gel electrophoresis was employed to analyze the protein patterns of (P.simoniiX P.nigra) XP.15 ACL tissue culture leaves in the different periods. An optimum method for 2-dimensional gel electrophoresis of leaves was set up. It was used to get better 2-DE images. By the ImageMaster diagram analysis software, we acquire a lot of protein points of obvious differences in expresses quality-protein point add, disappear etc. and measure- protein point dying deep or shallow. Most of them are proteins with low content and low molecular mass, which were probably related to the formation of adventitious roots primordium inducing, formation , individing and the further growth of adventitious roots respectively. Selecting 5 special expression protein points from the different periods 2-DE images that we proceed MALDI-TOF-MS measurese their PMF and set up the parameters to index in the database. Founding they are the relevant protein of growing and developing, material metabolizing ,cell factor, signal Conducting such as the SDE1, RIP, ATsEH etc.Predicting these protein points areclosely related with the formation, individing and further growth process of adventitious rootprimordium.
