In this study, mutant Populus tomentosa carr and wild type Populus tomentosa carr as test material, proteomics methods were used to explore the adventitious rooting molecular mechanisms of mutant Populus tomentosa carr cuttings.The main results were as following:1.Through different-expression protein analysis during adventitious rooting of mutant Populus tomentosa carr and Populus tomentosa carr cuttings,14 differentially expressed proteins were identified, among which 10 proteins were up-regulated and 4 proteins were down-regulated, between the three of mutant Populus tomentosa carr rooting inhibited, Populus tomentosa carr rooting and mutant Populus tomentosa carr rooting induced cuttings. Mass spectrometry and relational databases indicate that above proteins were bark storage protein, Chlorophyll a/b binding protein,60S acidic ribosomal protein P2 and unknown proteins.2.Three key periods of mutant Populus tomentosa carr adventitious root development were differentiation of primitive roots, from day 0 to days 3, root primordium formation, from days 3 to days 6, adventitious root elongation, from days 6 to days 12. In the three periods of mutant Populus tomentosa carr cuttings,49 differentially expressed proteins were identified, among which 15 proteins were up-regulated and 34 proteins were down-regulated.10 proteins were up-regulated from day 0 to days 3,24 proteins were up-regulated from 3 to days 6,35 proteins were up-regulated from days 6 to days 12. Mass spectrometry and relational databases indicate that differentially expressed proteins were cell division protein, bark storage protein, temperature-induced lipocalin, belt and delta subunits of ATP synthase, chlorophyll a/b binding protein, ribulose-1,5-bisphosphate carboxylase/oxygenase, triosephosphate isomerase, phosphoribulokinase,20kDa chaperonin, calreticulin, peroxiredoxin, glutathione transferase, CuZn-superoxide dismutase, polyphenol oxidase, beta-1,3-glucanase, kunitz-type trypsin inhibitor. Above proteins were the main participant in various biological processes, such as cell division, material transportation, energy metabolism, photosynthetic activity, calvin cycle, protein synthesis, antioxidant and defense. The several aspects of research suggested the adventitious rooting molecular mechanisms of mutant Populus tomentosa carr.cuttings.(1) The ratio of carbon nitrogen compounds affected the adventitious roots of cutting.(2) Energy metabolism and photosynthetic activity were the adventitious root formation of cuttings.(3) Calreticulin maintained the cellular homeostasis of adventitious roots.(4) Antioxidant and defense related proteins protected the occurrence of adventitious roots.Above results of the study are speculated that the adventitious root development of mutant Populus tomentosa carr cuttings may be a fully complex biological processes, with a variety of related proteins and be completed by multiple biological pathways coordinately.
